چکیده

A solid-state nuclear magnetic resonance (NMR) method for the site-resolved identification of the secondary structure of solid peptides and proteins is presented. This technique exploits the correlation between the backbone conformation and the CR chemical shift anisotropies (CSA) of proteins. The 13CR CSAs are measured under fast magic-angle-spinning using a new sequence of sixteen 180° pulses with special timing to reintroduce the CSA interaction selectively. Quantitative values of the shielding anisotropies are determined from the magnetization decay, as demonstrated for several amino acids. To achieve high-resolution spectra, this CSA filter experiment is combined with 2D 15N-13C correlation spectroscopy. Applied to selectively and extensively 13C-labeled and uniformly 15N-labeled ubiquitin at the largest dephasing time, the 2D experiment yields a spectral pattern that corresponds primarily to R-helical residues. This agrees with the previous finding that helical residues have smaller CSAs than sheet residues. However, the quantitative CSA differences between the helical and sheet conformations are less pronounced than indicated by solution-state NMR. This CSA filter technique provides an efficient and site-resolved method for characterizing the secondary structure of extensively isotopically labeled proteins.

Single-step determination of protein substructures using dipolar couplings: aid to structural genomics.

The wealth of genomic data that has recently become available with completion of the sequencing of both the human and a variety of other genomes1 has created a need for rapid and efficient determination of three-dimensional structures of the corresponding proteins. So far, most effort in this so-called structural genomics project has focused on X-ray crystallography, but NMR also shows consider...

Solution NMR Measurement of Amide Proton Chemical Shift Anisotropy in 15N-Enriched Proteins. Correlation with Hydrogen Bond Length§

Cross-correlation between 15N-1H dipolar interactions and 1HN chemical shift anisotropy (CSA) gives rise to different relaxation rates of the doublet components of 1H-{15N} peptide backbone amides. Two schemes for quantitative measurement of this effect are described and demonstrated for samples of uniformly 15N-enriched ubiquitin and perdeuterated 15N-enriched HIV-1 protease. The degree of rel...

Solid-state and high-resolution liquid 119Sn NMR spectroscopy of some monomeric, two-coordinate low-valent tin compounds: very large chemical shift anisotropies.

High-resolution liquid- and solid-state 119Sn NMR spectroscopy was used to study the bonding environment in the series of monomeric, two-coordinate Sn(II) compounds of formula Sn(X)C6H3-2,6-Trip2 (X = Cl, Cr(eta 5-C5H5)(CO)3, t-Bu, Sn(Me)2C6H3-2,6-Trip2; Trip = C6H2-2,4,6-i-Pr3). The trends in the principal components of the chemical shift tensor extracted from the solid-state NMR data were con...

Ultrahigh resolution protein structures using NMR chemical shift tensors.

NMR chemical shift tensors (CSTs) in proteins, as well as their orientations, represent an important new restraint class for protein structure refinement and determination. Here, we present the first determination of both CST magnitudes and orientations for (13)Cα and (15)N (peptide backbone) groups in a protein, the β1 IgG binding domain of protein G from Streptococcus spp., GB1. Site-specific...
